| I. Background | |
| 白細(xì)胞免疫球蛋白樣受體 B (LILRB)家族成員是髓細(xì)胞活化的負(fù)調(diào)節(jié)因子。LILRBs 屬 于 I 型跨膜糖蛋白，為免疫抑制性受體，目前已知的 LILRBs 包括 LILRB1、LILRB2、 LILRB3、LILRB4、LILRB5。 LILRB2（也被稱作 ILT4）包含四個胞外 Ig 結(jié)構(gòu)域、一個跨膜結(jié)構(gòu)域和三個 ITIM 基序， LILRB2 主要由髓系細(xì)胞表達(dá)，并作為 MDSCs 的主要抑制性受體。LILRB2 主要在嗜堿性 粒細(xì)胞、單核細(xì)胞、巨噬細(xì)胞、DC 和造血干細(xì)胞中表達(dá)。LILRB2 也表達(dá)于內(nèi)皮細(xì)胞、蛻 膜巨噬細(xì)胞，破骨細(xì)胞和神經(jīng)細(xì)胞。 LILRB2 的配體包括Ⅰ類蛋白 CD1c 和 CD1d、血管生成素樣蛋白、Nogo 受體配體和β- 淀粉樣蛋白。結(jié)合之后，LILRB2 通過 ITIM 基序與 SHP-1 磷酸酶的關(guān)聯(lián)來轉(zhuǎn)導(dǎo)負(fù)信號。 | |
